    A novel Flp reporter mouse shows that TRPA1 expression is largely limited to sensory neuron subsets | eNeuro
    Transient receptor potential ankyrin 1 (TRPA1) is a polymodal cation channel that is activated by electrophilic irritants, oxidative stress, cold temperature and GPCR signaling. TRPA1 expression has been primarily identified in subsets of nociceptive sensory afferents and is considered a target for future analgesics. Nevertheless, TRPA1 has been implicated in other cell types including keratinocytes, epithelium, enterochromaffin cells, endothelium, astrocytes and CNS neurons. Here, we developed a knock-in mouse which expresses the recombinase FlpO in TRPA1-expressing cells. We crossed the TRPA1Flp mouse with the R26ai65f mouse that expresses tdTomato in a Flp-sensitive manner. We found tdTomato expression correlated well with TRPA1 mRNA expression and sensitivity to TRPA1 agonists in subsets of TRPV1-expressing neurons in the vagal ganglia and dorsal root ganglia (DRG), although tdTomato expression efficiency was limited in DRG.     The role of perineuronal nets in the contralateral hemisphere in the electroacupuncture-mediated rehabilitation of post-stroke dysphagia mice | eNeuro
    Acupuncture at Lianquan (CV23) acupoint has been shown to improve swallowing function in post-stroke dysphagia (PSD). This improvement is supposed to be associated with the regulation of neuronal activity in the contralateral primary motor cortex (M1), while the underlying mechanism still needs to be elucidated. Perineuronal nets (PNNs) are well-known to be involved in the regulation of neuronal activity. Thus, we here aimed to detect the role of PNNs in the contralateral M1 hemisphere in the electroacupuncture (EA)-mediated effect in male mice. The results were obtained from a combination of methods, including in vitro slice electrophysiological recording, in vivo electrophysiological recording, and immunofluorescent staining in male mice.     Functional dissection of ipsilateral and contralateral neural activity propagation using voltage-sensitive dye imaging in mouse prefrontal cortex | eNeuro
    The prefrontal cortex (PFC) intrahemispheric activity and the interhemispheric connection significantly impacts neuropsychiatric disorder pathology This study aimed to generate a functional map of the PFC intra- and interhemispheric connections. Functional dissection of mouse PFCs was performed using the voltage-sensitive dye (VSD) imaging method with high speed (1 ms/frame), high resolution (256 × 256 pixels), and a large field of view (ca. 10 mm). Acute serial 350-µm slices were prepared from the bregma covering the PFC and numbered 1–5 based on their distance from the bregma (i.e., 1.70, 1.34, 0.98, 0.62, and 0.26 mm) with reference to the Mouse Brain Atlas. The neural response to electrical stimulation was measured at nine sites and then averaged; a functional map of the propagation patterns was created.     SaLSa: a combinatory approach of semi-automatic labeling and long short-term memory to classify behavioral syllables | eNeuro
    Accurately and quantitatively describing mouse behavior is an important area. Although advances in machine learning have made it possible to track their behaviors accurately, reliable classification of behavioral sequences or syllables remains a challenge. In this study, we present a novel machine learning approach, called SaLSa (a combination of semi-automatic labeling and long short-term memory-based classification), to classify behavioral syllables of mice exploring an open field. This approach consists of two major steps: first, after tracking multiple body parts, spatial and temporal features of their egocentric coordinates are extracted. A fully automated unsupervised process identifies candidates for behavioral syllables, followed by manual labeling of behavioral syllables using a graphical user interface.     Pupil-size sensitivity to listening demand depends on motivational state | eNeuro
    Motivation plays a role when a listener needs to understand speech under acoustically demanding conditions. Previous work has demonstrated pupil-linked arousal being sensitive to both listening demands and motivational state during listening. It is less clear how motivational state affects the temporal evolution of the pupil size and its relation to subsequent behavior. We used an auditory gap-detection task (N=33) to study the joint impact of listening demand and motivational state on the pupil-size response and examine its temporal evolution. Task difficulty and a listener’s motivational state were orthogonally manipulated through changes in gap duration and monetary-reward prospect. We show that participants’ performance decreased with task difficulty, but that reward prospect enhanced performance under hard listening conditions.     High-THC Cannabis smoke impairs incidental memory capacity in spontaneous tests of novelty preference for objects and odors in male rats | eNeuro
    Working memory is an executive function that orchestrates the use of limited amounts of information, referred to as working memory capacity, in cognitive functions. Cannabis exposure impairs working memory in humans; however, it is unclear if Cannabis facilitates or impairs rodent working memory and working memory capacity. The conflicting literature in rodent models may be at least partly due to the use of drug exposure paradigms that do not closely mirror patterns of human Cannabis use. Here, we used an incidental memory capacity paradigm where a novelty preference is assessed after a short delay in spontaneous recognition-based tests.     Origin of discrete and continuous dark noise in rod photoreceptors | eNeuro
    The detection of a single photon by a rod photoreceptor is limited by two sources of physiological noise, called discrete and continuous noise. Discrete noise occurs as intermittent current deflections with a waveform very similar to that of the single-photon response to real light and is thought to be produced by spontaneous activation of rhodopsin. Continuous noise occurs as random and continuous fluctuations in outer-segment current and is usually attributed to some intermediate in the phototransduction cascade. To confirm the origin of these noise sources, we have recorded from retinas of mouse lines with rods having reduced levels of rhodopsin, transducin or phosphodiesterase. We show that the rate of discrete noise is diminished in proportion to the decrease in rhodopsin concentration, and that continuous noise is independent of transducin concentration but clearly elevated when the level of phosphodiesterase is reduced.     The Precision of Place Fields Governs Their Fate Across Epochs of Experience | eNeuro
    Spatial memories are represented by hippocampal place cells during navigation. This spatial code is dynamic, undergoing changes across time – known as representational drift – and across changes in internal state, even while navigating the same spatial environment with consistent behavior. A dynamic code may provide the hippocampus a means to track distinct epochs of experience that occur at different times or during different internal states and update spatial memories.     Repeated exposure to high-THC Cannabis smoke during gestation alters sex ratio, behavior, and amygdala gene expression of Sprague Dawley rat offspring | eNeuro
    Due to the legalization of Cannabis in many jurisdictions and trend of increasing THC content in Cannabis products, an urgent need exists to understand the impact of Cannabis use during pregnancy on fetal neurodevelopment and behavior. To this end, we exposed female Sprague-Dawley rats to Cannabis smoke daily from gestational days 6 to 20 or room-air. Maternal reproductive parameters, offspring behavior, and gene expression in the offspring amygdala were assessed. Body temperature was decreased in dams following smoke exposure and more fecal boli were observed in the chambers before and after smoke exposure in dams exposed to smoke. Maternal weight gain, food intake, gestational length, litter number, and litter weight were not altered by exposure to Cannabis smoke.     Elevated serotonin in mouse spinal dorsal horn is pronociceptive | eNeuro
    Serotonergic neurons in the rostral ventral medulla (RVM) contribute to bidirectional control of pain through modulation of spinal and trigeminal nociceptive networks. Deficits in this pathway are believed to contribute to pathological pain states, but whether changes in serotonergic mechanisms are pro or anti-nociceptive is debated. We used a combination of optogenetics and fiber photometry to examine these mechanisms more closely. We find that optogenetic activation of RVM serotonergic afferents in the spinal cord of naïve mice produces mechanical hypersensitivity and conditioned place aversion.
