Journal ArticleNicotine robustly sustains smoking behavior by acting as a primary reinforcer and by enhancing the incentive salience of the nicotine-associated stimuli. The motivational effects produced by environmental cues associated with nicotine delivery can progressively manifest during abstinence resulting in reinstatement of nicotine seeking. However, how the activity in reward neuronal circuits is transformed during abstinence-induced nicotine seeking is not yet fully understood. In here we used a contingent nicotine and saline control self-administration model to disentangle the contribution of cue-elicited seeking responding for nicotine after drug abstinence in male Wistar rats. Journal ArticleCrisis? What crisis? Is the lack of reproducibility a supertramp that disperses widely across science fields? Is it carelessness, or is it also intrinsic to life sciences as I shall argue? Lack of reproducibility is a big flag brandished by many, but what do they mean by it? Where does it originate from? These were the first questions that crossed my mind as, while in Liège, Belgium for a PhD defense, I was asked to give a talk on the reproducibility crisis. Rather than a formal presentation, we had an open discussion. This editorial is its outcome. According to Wikipedia’s definition of reproducibility, “results obtained by an experiment or an observational study or in a statistical analysis of a data set should be achieved again with a high degree of reliability when the study is replicated” (https://en.wikipedia.org/wiki/Reproducibility). Journal ArticleMicroglia invade the neuroblast migratory corridor of the rostral migratory stream (RMS) early in development. The early postnatal RMS does not yet have the dense astrocyte and vascular scaffold that helps propel forward migrating neuroblasts, which led us to consider whether microglia help regulate conditions permissive to neuroblast migration in the RMS. GFP-labeled microglia in CX3CR-1GFP/+ mice assemble primarily along the outer borders of the RMS during the first postnatal week, where they exhibit predominantly an ameboid morphology and associate with migrating neuroblasts. Microglia ablation for 3 d postnatally does not impact the density of pulse labeled BrdU+ neuroblasts nor the distance migrated by tdTomato electroporated neuroblasts in the RMS. Journal ArticleFear is an emotional mechanism that helps to cope with potential hazards. However, when fear is generalized, it becomes maladaptive and represents a core symptom of posttraumatic stress disorder (PTSD). Converging lines of research show that dysfunction of glutamatergic neurotransmission is a cardinal feature of trauma and stress related disorders such as PTSD. However, the involvement of glutamatergic co-transmission in fear is less well understood. Glutamate is accumulated into synaptic vesicles by vesicular glutamate transporters (VGLUTs). The atypical subtype, VGLUT3, is responsible for the co-transmission of glutamate with acetylcholine, serotonin, or GABA. To understand the involvement of VGLUT3-dependent co-transmission in aversive memories, we used a Pavlovian fear conditioning paradigm in VGLUT3–/– mice. Journal ArticleMorphologically similar axon boutons form synaptic contacts with diverse types of postsynaptic cells. However, it is less known to what extent the local axonal excitability, presynaptic action potentials (APs), and AP-evoked calcium influx contribute to the functional diversity of synapses and neuronal activity. This is particularly interesting in synapses that contact cell types that show only subtle cellular differences but fulfill completely different physiological functions. Journal ArticleHippocampal neuronal activity at a time preceding stimulus onset affects episodic memory performance. We hypothesized that neuronal activity preceding an event supports successful memory formation; therefore, we explored whether a characterized encoding-associated brain activity, viz. the neuronal activity preceding a stimulus, predicts subsequent memory formation. To address this issue, we assessed the activity of single neurons recorded from the hippocampus in humans, while participants performed word memory tasks. Human hippocampal single-unit activity elicited by a fixation cue preceding words increased the firing rates (FRs) and predicted whether the words are recalled in a subsequent memory test; this indicated that successful memory formation in humans can be predicted by a preceding stimulus activity during encoding. Journal ArticleAllopregnanolone (AlloP) is a neurosteroid that potentiates ionotropic GABAergic (GABAA) inhibition and is approved for treating postpartum depression in women. Although the antidepressant mechanism of AlloP is largely unknown, it could involve selective action at GABAA receptors containing the δ subunit. Despite previous evidence for selective effects of AlloP on α4/δ-containing receptors of hippocampal dentate granule cells (DGCs), other recent results failed to demonstrate selectivity at these receptors (Lu et al., 2020). Journal ArticleScience is changing: the volume and complexity of data are increasing, the number of studies is growing and the goal of achieving reproducible results requires new solutions for scientific data management. In the field of neuroscience, the German National Research Data Infrastructure (NFDI-Neuro) initiative aims to develop sustainable solutions for research data management (RDM). To obtain an understanding of the present RDM situation in the neuroscience community, NFDI-Neuro conducted a comprehensive survey among the neuroscience community. Here, we report and analyze the results of the survey. We focused the survey and our analysis on current needs, challenges, and opinions about RDM. Over the past decades, several animal models of focal cerebral ischemia have been developed allowing to investigate pathophysiological mechanisms underlying stroke progression. Despite intense preclinical research efforts, the need for noninvasive mouse models of vascular occlusion targeting the middle cerebral artery yet avoiding mechanical intervention is still pressing. Here, by applying the photothrombotic stroke model to the distal branch of the middle cerebral artery, we developed a novel strategy to induce a targeted occlusion of a large blood vessel in mice. This approach induces unilateral damage encompassing most of the dorsal cortex from the motor up to the visual regions 1 week after stroke. Neuroimaging studies based on functional magnetic resonance imaging (fMRI) identified specific characteristics of spontaneous (intrinsic) brain dynamics to be associated with individual differences in general cognitive ability, i.e., intelligence. However, fMRI research is inherently limited by low temporal resolution, thus, preventing conclusions about neural fluctuations within the range of milliseconds.
